One thing that resonates from the two-day XMRV conference at the
National Institutes of Health is the, at times, testy battle between two
camps of scientists. The cohort that believes that mouse retrovirus
contamination accounts for the positive XMRV and related murine leukemia
virus studies was pitted against the cohort that believes contamination
has been ruled out and that the positive studies show a clear link
between this family of retroviruses and Chronic Fatigue Syndrome and
prostate cancer. |

Researchers have found the presence of a retrovirus called XMRV in blood
samples from a high percentage of Chronic Fatigue (ME/CFS) patients who
took part in the study. A small number
of Fibromyalgia samples also tested positive for XMRV. It is not yet clear
whether XMRV is the underlying cause of

In collaboration with the laboratories of Judy Mikovits and Frank Ruscetti,
we were able to use antibodies developed against the envelope protein of
SFFV to detect infectious XMRV in the blood cells and plasma of patients
suffering from the neuroimmune disease chronic fatigue syndrome (CFS). We
were further able to develop a seroconversion assay using cells expressing
the SFFV envelope protein to detect antibodies against the virus in the
plasma of CFS patients. We now plan to apply our knowledge of the pathogenesis
of mouse retroviruses that cause cancer and neurological disease in rodents
to study the molecular basis for similar diseases associated with XMRV.

Identified DNA
from a human gammaretrovirus, xenotropic murine leukemia virus-related virus
(XMRV), in 68 of 101 patients (67%) compared to 8 of 218 (3.7%) healthy
controls. Cell culture experiments revealed that patient-derived XMRV is
infectious and that both cell-associated and cell-free transmission of the
virus are possible. Secondary viral infections were established in uninfected
primary lymphocytes and indicator cell lines following exposure to activated
PBMCs, B cells, T cells, or plasma derived from CFS patients. These findings
raise the possibility that XMRV may be a contributing factor in the pathogenesis
of CFS.

How is the virus
transmitted? Is XMRC by itself a symptom-causing disease, in addition to
the peculiar role it plays in conjunction with other diseases (as do all
retroviruses)? What diseases are activated by XMRC? What treatments are

XMRV is a gammaretrovirus, one of a family of viruses long-studied in animals
but not known to infect people. In animals, these retroviruses can cause
horrendous neurological problems, immune deficiency, lymphoma and leukemia.
The new study provided overwhelming evidence that XMRV is a human gammaretrovirus
— the third human retrovirus (after H.I.V. and human lymphotropic viruses,
which cause leukemia and lymphoma). Infection is permanent and, yes, it
can spread from person to person (though it is not yet known how the virus
is transmitted
